Effects of lipopolysaccharide exposure on the inflammatory response, butyrate flux, and metabolic function of the ruminal epithelium using an ex vivo model

Acidotic conditions in the rumen have been associated with compromised barrier function of ruminal epithelium and translocation microbe-associated molecular patterns (MAMP) such as lipopolysaccharide (LPS). Interaction MAMP may also induce a local proinflammatory response. The aim this study was to evaluate potential response following LPS exposure Ussing chambers, investigate whether affects flux metabolism butyrate. Ruminal epithelial tissue from 9 Holstein bull calves were mounted into chambers exposed 0, 10,000, 50,000, or 200,000 endotoxin units (EU)/mL for duration 5 h. Radiolabeled 14C-butyrate (15 mM) added mucosal buffer assess mucosal-to-serosal 14C-butyrate. Additional without radioisotope, either 0 EU/mL used measure release ?-hydroxybutyrate (BHB) IL1B buffer, collect analysis gene expression. Butyrate not significantly affected by exposure; however, we detected tendency butyrate increase linearly dose. Bidirectional releases BHB exposure. Expression TLR4, MCU downregulated ex vivo. We no effects on expression genes transport. results present are interpreted indicate that, although inflammatory slightly suppressed, altered function.
